Cell Line Description P815 is a murine mastocytoma cell line derived from a methylcholanthrene-induced tumor in DBA/2 mice. P815 cells exhibit a lymphoblast-like or round morphology and grow in suspension culture as single cells or loose aggregates. The cell line retains key biological characteristics of mast cells, including the expression of Fcε receptors and the capacity for histamine synthesis. P815 is highly tumorigenic in syngeneic DBA/2 mice, making it one of the most widely used immunocompetent murine tumor models in cellular immunology. Due to the expression of well-defined tumor-associated antigens (such as P1A), P815 serves as a fundamental in vitro and in vivo model for studying cytotoxic T lymphocyte (CTL) responses, tumor antigen presentation, and mechanisms of immune escape; it is also extensively used to evaluate cancer immunotherapies, cell-mediated cytotoxicity assays, and cancer vaccines.

Culture Conditions and Handling
Subculturing 1. Maintain the culture by adding fresh medium or replacing the medium.
2. Establish the culture by centrifuging (1,000 rpm, 5 minutes) and resuspending the cells (at a density of 1 × 10⁵ to 2 × 10⁵ viable cells/mL).
3. Do not allow the cell concentration to exceed 1 × 10⁶ cells/mL.
4. Gently resuspend the cells to prepare a single-cell suspension before subculturing into a new culture vessel.

Subcultivation Ratio Maintain cell density between 1 × 10⁵ and 1 × 10⁶ cells/mL (split ratio approximately 1:3 to 1:6)
