HC11 Cell Line

Cell Line Description HC11 is a clonal mouse mammary epithelial cell line derived from the COMMA-D cell line (which originated from mid-pregnant BALB/c mouse mammary tissue). HC11 cells exhibit an epithelial morphology and grow as an adherent monolayer under standard in vitro culture conditions. A distinctive feature of this cell line is its retained capacity for functional differentiation in response to lactogenic hormones; upon combined induction with prolactin, dexamethasone, and insulin, HC11 cells rapidly differentiate and express milk protein genes, particularly the β-casein gene. Genetically, the cell line harbors a point mutation in the Trp53 gene that leads to immortalization, yet it maintains near-normal, non-tumorigenic biological characteristics under standard conditions. HC11 serves as a highly valuable and physiologically relevant model for studying mammary gland development, lactogenic signaling (such as the STAT5 pathway), mechanisms of hormone-regulated gene expression, and early events in mammary tumorigenesis.

Culture Conditions and Handling
Subculturing 1. Remove and discard the culture medium.
2. Briefly rinse the cell monolayer with PBS (without Ca2+/Mg2+) to completely remove residual serum.
3. Add 1.0 to 2.0 mL of 0.25% trypsin-0.03% EDTA solution and incubate at 37°C until the cell monolayer detaches (typically 2 to 5 minutes).
4. Add fresh complete growth medium to neutralize the trypsin and gently pipette up and down to prepare a single-cell suspension.
5. Subculture the cell suspension into new culture vessels.
