Cell Line DescriptionThe DT40 chicken B cell line is derived from avian leukemia virus (ALV)-induced bursa lymphoma. ALV was injected intravenously into Hyline SC chickens, and the resulting tumors were transplanted twice in vivo before cell culture. DT40 cells are small, approximately 10 μm in diameter, and have a high nuclear-to-cytoplasmic ratio. They carry IgM on their surface. DT40 cells do release ALV into the surrounding environment, but in negligible amounts. The DT40 cell line is a versatile system that can be used to study specific signal transduction pathways, such as those activated by BCR or induced by apoptotic factors, as well as the study of nucleic acid modification, repair, and tissue. DT40 is easy to culture, and mutations in Ig genes can be easily measured by using FACS to quantify the loss of surface immunoglobulins. Furthermore, gene targeting efficiency in DT40 was very high, possibly due to elevated levels of enzymes involved in homologous recombination for gene conversion. This makes DT40 a useful model for analyzing gene function.

It provides a targeted DNA to random DNA integration ratio of over 1:2, far exceeding that of any mammalian cell line.
