MDBK Cell Line

Cell Line Description MDBK is a continuous bovine kidney epithelial cell line established by S.H. Madin and N.B. Darby in 1957. The cell line was derived from normal kidney tissue of a healthy adult steer (a castrated male bovine). MDBK cells grow as an adherent monolayer with tight intercellular junctions, exhibiting a characteristic "cobblestone" epithelial morphology. MDBK is recognized as a cornerstone model system in the fields of veterinary medicine, virology, and biotechnology. It is widely used for the isolation, diagnostic propagation, and industrial-scale vaccine production of various important veterinary viruses, particularly bovine viral diarrhea virus (BVDV) and bovine herpesvirus type 1 (BHV-1). Additionally, MDBK serves as a common non-human host system for assaying interferon activity and studying viral replication kinetics.

Characteristics
Tumorigenic No, non-tumorigenic under standard physiological baselines. Does not form tumors when engrafted into standard immunocompromised rodent hosts.

Viral Sensitivity Status It is naturally highly susceptible to a variety of viruses, including bovine herpesvirus type 1, bovine viral diarrhea virus (BVDV), vesicular stomatitis virus (Indiana strain), vaccinia virus, and adenovirus.
Growth Kinetics Rapid, highly robust adherent proliferation; typical population doubling time spans approximately 20 to 30 hours.

Culture Conditions and Handling
Subculturing 1. Remove and discard all used complete culture medium from the culture dish.
2. Gently rinse the confluent cell layer with sterile PBS or a 0.25% trypsin-0.53 mM EDTA solution (Ca2+/Mg2+-free) to neutralize residual serum.
3. Aspirate the rinse solution, add 2.0 to 3.0 mL of fresh trypsin-EDTA solution to cover the cell layer, and let it sit.
4. Incubate at 37°C for 2 to 7 minutes; monitor closely under an inverted microscope until the epithelial cell layer completely detaches, rounds up, and becomes mobile.
5. Add 6.0 to 8.0 mL of pre-warmed complete culture medium to the dish and gently pipette the cells up and down to achieve a uniform single-cell suspension.
6. Centrifuge at approximately 125 × g for 5 minutes, discard the supernatant, resuspend the cell pellet uniformly in fresh complete culture medium, and distribute the suspension into new vessels.
Medium Renewal 2 to 3 times per week
Subcultivation Ratio Split optimal, confluent cultures at a standard ratio of 1:4 to 1:10. This line grows rapidly and tolerates high dilution steps easily.
