CaSki Cell Line

General Information
Organism Homo sapiens, human
Cell Line Description CaSki is a human cervical epidermoid carcinoma cell line, derived from a small intestine metastasis in a 40-year-old female patient with cervical cancer. CaSki cells exhibit an epithelial morphology and grow as an adherent monolayer in culture. A key molecular feature of this cell line is the presence of an integrated human papillomavirus type 16 (HPV-16) genome—approximately 600 copies per cell—along with sequences related to HPV-18; the cells also express the HPV E6 and E7 oncoproteins. Cytogenetically, the cell line displays an aneuploid karyotype characterized by various complex chromosomal alterations. Due to the integration of the endogenous HPV-16 genome and the high-level expression of viral oncogenes, CaSki has become a classic and widely utilized in vitro model. It is frequently employed to investigate the pathogenesis of cervical cancer, explore the mechanisms of HPV-mediated cellular transformation, evaluate antiviral and targeted antitumor therapies, and test prophylactic or therapeutic HPV vaccines.

Culture Conditions and Handling
Subculturing 1. Remove and discard the culture medium.
2. Briefly rinse the cell monolayer with PBS (without Ca2+/Mg2+) to completely remove residual serum.
3. Add 1.0 to 2.0 mL of 0.25% trypsin-0.03% EDTA solution and incubate at 37°C until the cell monolayer detaches (typically 2 to 5 minutes).
4. Add fresh complete growth medium to neutralize the trypsin and gently pipette up and down to prepare a single-cell suspension.
5. Subculture the cell suspension into new culture vessels.
Medium Renewal Every 2 to 3 days
Subcultivation Ratio A split ratio of 1:3 to 1:6 is recommended
Culture Conditions Atmosphere: Air, 95%; CO2, 5%; Temperature: 37°C
Cryopreservation Complete growth medium supplemented with 5% to 10% (v/v) DMSO
