MPC-5 Cell Line

Cell Line Description MPC-5 is a conditionally immortalized mouse podocyte cell line established by P. Mundel and colleagues in 1997. This cell line was isolated from the kidneys of H2Kb-tsA58 transgenic mice (the "Immortomouse" background) carrying the temperature-sensitive simian virus 40 large T antigen (SV40LT), which is controlled by an interferon-γ (IFN-γ) inducible promoter. This unique genetic system allows MPC-5 to switch between a proliferating undifferentiated state and a non-proliferating mature podocyte state. Due to this dual nature, MPC-5 has become the global gold standard in vitro model for studying glomerular visceral epithelial biology, podocyte differentiation, slit membrane molecular dynamics, and the mechanisms of damage associated with diabetic nephropathy and focal segmental glomerulosclerosis (FSGS).

Growth States 1. Permissive culture (33°C + IFN-γ): Rapid proliferation, pebble-like morphology, SV40LT active expression.
2. Restrictive culture (37°C, without IFN-γ): Growth arrest, extensive dendritic/foot process expansion, SV40LT degradation.
Expression Markers WT1 (Wilms' Tumor 1) was constitutively expressed in both states; synaptoprotein, renin, podocyte protein and CD2AP were significantly upregulated 10-14 days after differentiation at 37°C.

Culture Conditions and Handling
Subculturing (Expansion Only) 1. Remove and discard the culture supernatant.
2. Gently wash the cell monolayer with Ca²⁺/Mg²⁺-free PBS.
3. Add 0.25% trypsin-0.53 mM EDTA solution and incubate at 33°C (usually 3 to 6 minutes) until cells detach.
4. Neutralize the trypsin with complete culture medium; gently pipette to obtain a single-cell suspension.
5. Transfer the cells to a new culture dish. Note: Subculturing for maintenance and expansion of cells must be performed at 33°C and with the addition of IFN-γ.
Differentiation Induction To initiate the differentiation process for subsequent experiments, cells were collected at 33°C, resuspended in complete medium completely free of IFN-γ, and seeded onto experimental culture plates. Cells were strictly cultured at 37°C for 10 to 14 days without subculturing until a mature differentiation phenotype was formed.
Subcultivation Ratio A split ratio of 1:3 to 1:6 is recommended (during the 33°C expansion phase)
Culture Conditions Atmosphere: Air, 95%; CO2, 5%; Temperature: 33°C for growth, 37°C for differentiation.
Cryopreservation 90% complete culture medium (containing IFN-γ) + 10% DMSO
