BT-20 Cell Line

General Information
Organism Homo sapiens, human
Cell Line Description BT-20 is a human breast adenocarcinoma cell line established by E.Y. Lasfargues and L. Ozzello in 1958, derived from a primary breast tumor in a 74-year-old Caucasian female. It is one of the earliest and most widely used models for triple-negative breast cancer (TNBC). The cell line is negative for the estrogen receptor (ER), progesterone receptor (PR), and HER2, though it expresses an ER mRNA variant lacking exon 5. These cells grow as an adherent epithelial monolayer with a doubling time of approximately 66 to 70 hours and can form grade II adenocarcinomas in nude mice. BT-20 harbors a homozygous TP53 p.Lys132Gln mutation, two heterozygous PIK3CA mutations, and a homozygous CDKN2A deletion; furthermore, the cell line exhibits microsatellite stability. It is extensively utilized in research concerning TNBC biology, apoptosis and TRAIL sensitivity, EGFR and PI3K signaling pathways, and three-dimensional spheroid culture.

Culture Conditions and Handling
Subculturing 1. Remove and discard the culture medium.
2. Briefly rinse the cell monolayer with PBS (Ca²⁺/Mg²⁺-free) to thoroughly remove residual serum.
3. Add 2.0 to 3.0 mL of 0.25% Trypsin–0.53 mM EDTA solution, and observe under an inverted microscope until the cell monolayer has dispersed (typically takes 2 to 5 minutes).
4. Add 6.0 to 8.0 mL of complete growth medium, and gently pipette to prepare a single-cell suspension.
5. Aliquot the cell suspension into new culture vessels.
Medium Renewal 2 to 3 times per week
Subcultivation Ratio A split ratio of 1:2 to 1:4 is recommended.
Culture Conditions Atmosphere: Air, 95%; CO2, 5%; Temperature: 37°C
Cryopreservation Complete growth medium supplemented with 5% (v/v) DMSO
