Caki-1 Cell Line

General Information
Organism Homo sapiens, human
Cell Line Description Caki-1 is a human renal cell carcinoma cell line derived from a cutaneous metastasis in a 49-year-old Caucasian male patient with clear cell renal cell carcinoma (ccRCC). Caki-1 cells exhibit an epithelial morphology and grow as an adherent monolayer under standard culture conditions. The cell line retains distinct ultrastructural characteristics of proximal renal tubular epithelial cells, such as microvilli, multilayered cellular lamellae, and microfilaments. Cytogenetically, Caki-1 displays an aneuploid karyotype with complex chromosomal alterations. At the molecular level, Caki-1 expresses wild-type VHL, making it a valuable model for comparative studies with VHL-mutant renal cancer cell lines. Furthermore, the cell line demonstrates high tumorigenicity and metastatic potential in vivo. Due to its well-defined phenotypic and physiological properties, Caki-1 is widely used as an in vitro model to investigate the biology of clear cell renal carcinoma, explore angiogenesis and hypoxia pathways, evaluate anti-angiogenic drugs and targeted therapies, and conduct functional genomic screens.

Culture Conditions and Handling
Subculturing 1. Remove and discard the culture medium.
2. Briefly rinse the cell monolayer with PBS (without Ca2+/Mg2+) to completely remove residual serum.
3. Add 1.0 to 2.0 mL of 0.25% trypsin-0.03% EDTA solution and incubate at 37°C until the cell monolayer detaches (typically 2 to 5 minutes).
4. Add fresh complete growth medium to neutralize the trypsin and gently pipette up and down to prepare a single-cell suspension.
5. Subculture the cell suspension into new culture vessels.
Medium Renewal Every 2 to 3 days
Subcultivation Ratio A split ratio of 1:3 to 1:6 is recommended
Culture Conditions Atmosphere: Air, 95%; CO2, 5%; Temperature: 37°C
Cryopreservation Complete growth medium supplemented with 5% to 10% (v/v) DMSO
