Cell Line Description SW13 is a human adrenocortical carcinoma cell line established by A. Leibovitz and his colleagues in 1971. This cell line was isolated from the primary tumor tissue of a 55-year-old female patient diagnosed with small-cell adrenocortical carcinoma. SW13 cells exhibit a small, epithelial-like morphology and grow as adherent monolayers. This cell line holds immense significance in the fields of modern oncology and cell biology, as it is widely recognized as the global "gold standard" model for studying defects in the SWI/SNF chromatin remodeling complex. Due to its inherent deficiency in the core subunits of the SWI/SNF complex—specifically the proteins BRG1 (SMARCA4) and BRM (SMARCA2)—SW13 cells are extensively utilized to investigate epigenetic gene regulation, defects in steroid biosynthesis, and viral replication mechanisms (such as the proliferation processes of influenza viruses, polyomaviruses, and flaviviruses).

Tumorigenic Yes, reliably forms small-cell carcinomas when engrafted into immunocompromised mice (e.g., nude or SCID mice)

Genetic Profile 1. BRG1 (SMARCA4) Deficiency: Exhibits a constitutive state of deficiency resulting from genomic mutation or silencing.

3. Vimentin Expression: Displays cellular heterogeneity, manifesting as distinct vimentin-positive and vimentin-negative subclones, both of which exhibit divergent structural dynamic characteristics.
Growth Kinetics Highly proliferative and dense monolayer expansion; doubling time typically ranges between 24 to 34 hours.

Culture Conditions and Handling
Subculturing 1. Completely remove and discard the spent culture medium.
2. Gently rinse the confluent cell monolayer with Ca²⁺/Mg²⁺-free Dulbecco's Phosphate-Buffered Saline (DPBS) to neutralize residual serum components.
3. Add 2.0 to 3.0 mL of 0.25% Trypsin–0.53 mM EDTA solution, ensuring the cell layer is completely submerged.
4. Incubate at 37°C for 2 to 6 minutes; observe under an inverted microscope until the cell layer has completely detached and the cells have rounded up.
5. Add an equal or greater volume of complete culture medium to thoroughly neutralize the trypsin activity.
6. Centrifuge at approximately 200-300 × g for 5 minutes; discard the supernatant, gently resuspend the cell pellet in fresh medium, and aliquot evenly into new culture flasks.
Medium Renewal 2 to 3 times per week
Subcultivation Ratio A split ratio of 1:3 to 1:6 is standardly recommended for routine maintenance.
