BHK21(C-13)

General Information
Organism Mesocricetus auratus, Syrian golden hamster
Cell Line Description BHK-21 clone 13 (BHK-21/C-13) is a fibroblast cell line established in 1961 by I.A. Macpherson and M.G.P. Stoker from the kidneys of one-day-old Syrian golden hamsters. Derived from the parental BHK-21 cell line, it was isolated via single-cell cloning following 84 days of continuous culture. Since then, it has become one of the most widely used cell lines in the fields of virology and biotechnology. BHK-21/C-13 grows as an adherent monolayer and exhibits a fibroblast-like morphology; it has a doubling time of approximately 25–50 hours and a pseudodiploid karyotype (2n=44). The cell line is highly susceptible to a variety of viruses—including rabies virus, foot-and-mouth disease virus, vesicular stomatitis virus, herpes simplex virus, and poliovirus—and is recommended by the World Organisation for Animal Health (OIE) for routine rabies diagnosis. Furthermore, BHK-21/C-13 is extensively utilized as a host for transfection in the production of recombinant proteins and viral vectors, in studies of cell transformation induced by chemical carcinogens, and as a platform for three-dimensional cell culture, high-throughput toxicology, and drug screening. Historically, it has also served as an important substrate for the development of live attenuated veterinary vaccines.

Characteristics
Doubling Time Approximately 25–50 hours
Tumorigenic Forms subcutaneous fibrosarcomas in weanling hamsters and in immunodeficient hosts following inoculation
Karyotype Pseudodiploid; 2n=44, with marker chromosomes 6q+ and 15q+ in the majority of cells. Approximately 4–21% tetraploid cells are observed
Genetic Profile Spontaneously immortalized fibroblast line; species identity confirmed by mitochondrial COI barcoding
Mycoplasma Test Negative
Culture Conditions and Handling
Subculturing 1. Remove and discard the culture medium.
2. Briefly rinse the cell monolayer with PBS (Ca²⁺/Mg²⁺-free) to thoroughly remove residual serum.
3. Add 2.0 to 3.0 mL of 0.25% Trypsin–0.53 mM EDTA solution, and observe under an inverted microscope until the cell monolayer has dispersed (typically takes 2 to 5 minutes).
4. Add 6.0 to 8.0 mL of complete growth medium, and gently pipette to prepare a single-cell suspension.
5. Aliquot the cell suspension into new culture vessels.
Medium Renewal 1 to 2 times per week
Subcultivation Ratio A split ratio of 1:2 to 1:10 is recommended; seed at approximately 2–4 × 10⁴ cells/cm².
Culture Conditions Atmosphere: Air, 95%; CO2, 5%; Temperature: 37°C
Cryopreservation Complete growth medium supplemented with 5% (v/v) DMSO, or 70% complete growth medium + 20% FBS + 10% DMSO
