Cell Line DescriptionHepa1-6 is a murine hepatocellular carcinoma cell line that is widely used in cancer research as a model system to study liver cancer biology. It was originally derived from spontaneous liver cancer in C57BL/6 mice, and its growth properties, gene expression profile, and response to various anticancer agents have been extensively characterized. These cells have an epithelial morphology and exhibit an undifferentiated hepatocellular carcinoma phenotype. Hepa 1-6 cells are known for their ability to be easily cultured and to maintain stable growth and propagation under standard laboratory conditions. Hepa1-6 cells are often used in preclinical studies to evaluate the efficacy and toxicity of novel anticancer drugs, as well as to study the mechanisms underlying tumor growth and drug resistance. They are also frequently used in immunology studies to study the interactions between cancer cells and immune cells and to develop immunotherapeutic approaches for cancer treatment. Hepa1-6 cells are often used to study the molecular mechanisms underlying liver cancer development and progression, as well as to evaluate the efficacy and toxicity of novel cancer therapies. Researchers also use Hepa1-6 cells to study the interactions between cancer cells and the immune system and to develop immunotherapeutic approaches for cancer treatment.

Additional InfoThe Hepa 1-6 cell line was used to create the cell line-derived (CDX) xenograft Hepa 1-6 mouse model. The Hepa 1-6 xenograft model can be used to study the inhibitory effects of therapeutic agents on tumor growth, such as recombinant adenoviral expression of hTERTC27 or RNA replacement therapy through miRNA regulation (i.e., miR-122 or miR-34).

Culture Conditions and Handling
SubculturingVolumes given are for 75 cm2 flasks. For culture vessels of other sizes, increase or decrease the amount of dissociation medium required proportionally.
1. Remove and discard the medium.
2. Rinse the cell layer briefly with 0.25% (w/v) trypsin - 0.53 mM EDTA solution to remove all traces of serum containing trypsin inhibitors.
3. Add 2.0 to 3.0 mL of trypsin-EDTA solution to the flask and observe the cells under an inverted microscope until the cell layer is dispersed (usually within 5 to 15 minutes). Note: To avoid clumping, do not agitate the cells by tapping or shaking the flask while waiting for the cells to detach. Cells that are difficult to detach can be placed at 37°C to facilitate dispersion.
4. Add 6.0 to 8.0 mL of complete growth medium and gently pipette out the cells.
5. Add an appropriate aliquot of the cell suspension to a new culture vessel.
6. Incubate the culture at 37°C.
