ZJU-0430 Cell Line

Cell Line Description The ZJU-0430 gallbladder cancer (GBC) cell line was derived from a 74-year-old male patient who presented with abnormal serum ferritin levels and whose gallbladder cancer was pathologically classified as a well-differentiated adenocarcinoma. Under microscopic examination, this cell line exhibits diverse cellular morphologies and typical epithelial cell characteristics; however, scRNA-Seq analysis revealed that its internal heterogeneity is not significant. Its population doubling time (PDT) is 19.81 hours, which is shorter than that of the GBC-SD cell line. Immunophenotypic analysis results indicate that ZJU-0430 cells highly express CD24, CD44, CD29, and CD133; partially express CD184 and CD326; and do not express CD34, CD90, CD117, or CD338. Compared to GBC-SD cells, ZJU-0430 cells demonstrate superior migratory, invasive, and proliferative capabilities. Furthermore, in a nude mouse xenograft model, these cells also exhibited in vivo tumorigenicity. Consequently, the ZJU-0430 cell line serves as a highly representative model for the disease and is suitable for preclinical studies investigating the pathogenesis and therapeutic strategies of gallbladder cancer.

Culture Conditions and Handling
Subculturing 1. Remove and discard the culture medium.
2. Briefly rinse the cell monolayer with PBS (Ca²⁺/Mg²⁺-free) to thoroughly remove residual serum.
3. Add 2.0 to 3.0 mL of 0.25% Trypsin–0.53 mM EDTA solution, and observe under an inverted microscope until the cell monolayer has dispersed (typically takes 5 to 10 minutes).
4. Add 6.0 to 8.0 mL of complete growth medium and gently pipette to prepare a single-cell suspension.
5. Aliquot the cell suspension into new culture vessels.
