| Cell Line Description |
KYSE-520 is a human esophageal squamous cell carcinoma (ESCC) cell line derived from an invasive, moderately differentiated tumor located in the lower thoracic esophagus of a 58-year-old Japanese female; the primary tumor had invaded the adventitia. As a member of the well-characterized KYSE series of esophageal cancer cell lines, KYSE-520 exhibits an epithelioid morphology and grows as an adherent monolayer, with giant cells occasionally observed within the population (accounting for approximately 1%). Genetically, this cell line harbors a splice acceptor site mutation (c.376-2A>T) in the tumor suppressor gene TP53, alongside an amplification of the oncogene MYC—both of which are frequently reported alterations in esophageal squamous cell carcinoma. KYSE-520 expresses various squamous and epithelial markers, including pan-cytokeratin, cytokeratins 8, 17, 18, and 19, as well as EpCAM and vimentin; however, it does not express cytokeratin 7, desmin, glial fibrillary acidic protein (GFAP), or neurofilament. This cell line serves as a practical platform for investigating the mechanisms of squamous cell carcinogenesis, p53-dependent signaling pathways, tumor radiosensitivity, and chemotherapy resistance, as well as for evaluating potential therapeutic agents. Given its well-defined genetic background, KYSE-520 is widely utilized in both monolayer cell assays and animal models of esophageal cancer, playing a significant role in target validation and biomarker discovery. |

| Culture Conditions and Handling |
| Subculturing |
1. Remove and discard the culture medium. 2. Briefly rinse the cell monolayer with PBS (Ca²⁺/Mg²⁺-free) to thoroughly remove residual serum. 3. Add 2.0 to 3.0 mL of 0.25% Trypsin–0.53 mM EDTA solution, and observe under an inverted microscope until the cell monolayer has dispersed (typically takes 2 to 5 minutes). 4. Add 6.0 to 8.0 mL of complete growth medium, and gently pipette to prepare a single-cell suspension. 5. Aliquot the cell suspension into new culture vessels. |
| Medium Renewal |
2 to 3 times per week |
| Subcultivation Ratio |
When seeding new culture vessels, the cell density should be approximately 0.5-1.0 X 10⁶ cells per 80 cm². Once the cells reach confluence, they should be subcultured using trypsin/EDTA at a split ratio of 1:3 to 1:6, with subculturing performed every 3–4 days (yielding approximately 4-5 X 10⁶ cells per 80 cm²). |
