| General Information |
| Organism |
Homo sapiens, human |
| Cell Line Description |
MFE-280 is a human endometrial adenocarcinoma cell line established from recurrent tumor tissue. The donor tumor was described as adenomatous, partly papillary, and high grade (G3). The cells grow as an adherent epithelial-like monolayer and have a relatively slow reported doubling time. Documented progesterone receptor expression, placental protein 14 expression, microsatellite stability, and defined PIK3CA and TP53 variants make this model useful for mechanistic and translational studies of high-grade endometrial cancer, hormone-related biology, oncogenic signaling, and therapeutic response. |

| Culture Conditions and Handling |
| Culture Conditions |
37°C in a humidified atmosphere containing 5% CO₂ |
| Recommended Seeding Density |
Seed 2–6 × 10⁶ cells per 75 cm² culture flask |
| Passage Endpoint |
Subculture when the monolayer is confluent |
| Subcultivation Ratio |
Split at a ratio of 1:2 to 1:6 every 3–5 days, using trypsin/EDTA for detachment |
| Subculturing |
1. Remove the spent medium from a confluent culture. 2. Detach the monolayer with trypsin/EDTA while monitoring the cells microscopically. 3. Add complete growth medium to stop dissociation and collect the cell suspension. 4. Resuspend gently to obtain an even suspension. 5. Seed fresh culture vessels at a 1:2 to 1:6 split ratio, maintaining the reported inoculum range where possible. 6. Return the cultures to 37°C and 5% CO₂ and monitor attachment and growth. |
| Post-Thaw Recovery |
Seed the thawed suspension into a 75 cm² flask using complete medium. Approximately 4 hours after seeding, transfer the medium and floating cells into a second flask rather than discarding them; this preserves the initially non-adherent fraction described in the referenced recovery procedure. Continue both cultures under the standard incubation conditions. |
| Cryopreservation Medium |
70% complete culture medium + 20% fetal bovine serum + 10% DMSO |
