PC-3M Cell Line

Cell Line Description PC-3M is a highly metastatic, adherent human prostate cancer cell line established in 1984 by J.M. Kozlowski and colleagues. It was isolated as a subline selected in vivo from liver metastases in nude mice after intrasplenic injection of a parental PC-3 cell line (originally derived from bone metastases in a 62-year-old white male). PC-3M cells grow as adherent monolayers of epithelial-like cells. PC-3M is globally recognized as the premier model for treating advanced invasive prostate cancer, exhibiting significantly increased invasiveness and metastatic potential compared to parental PC-3 cells. It is extensively used in cancer research to investigate tumor cell migration, extravasation, epithelial-mesenchymal transition (EMT), and to screen anti-metastatic therapies.

Characteristics
Tumorigenic Yes, highly tumorigenic and highly metastatic in nude mice, with a strong propensity to colonize the liver, lungs, and lymph nodes.

Culture Conditions and Handling
Subculturing 1. Completely remove and discard the used complete culture medium.
2. Gently wash the cell monolayer with sterile PBS (Ca2+/Mg2+-free) to remove any residual serum.
3. Add 1.0 to 2.0 mL of 0.25% trypsin-0.53 mM (0.03%) EDTA solution to completely cover the cell layer.
4. Incubate at room temperature or 37°C for 2 to 5 minutes; observe under an inverted microscope until the cells become rounded, loose, and completely detached.
5. Add an equal volume of complete culture medium to rapidly neutralize the trypsin activity.
6. Centrifuge the cell suspension at approximately 125 × g for 5 minutes. Carefully aspirate the supernatant, gently resuspend the pellet in fresh complete culture medium, and aliquot into new containers.
Medium Renewal 2 to 3 times per week
Subcultivation Ratio Split confluent cultures at a standard ratio of 1:3 to 1:8. Do not allow the cells to remain at 100% confluence for prolonged periods to prevent cell piling and shedding.
