Cell Line Description KLN205 is a murine lung squamous cell carcinoma cell line derived from a spontaneous lung tumor in a DBA/2 mouse. The cells exhibit an epithelial morphology and grow as an adherent monolayer under standard in vitro culture conditions. KLN205 cells retain key histological and cytological characteristics of lung squamous cell carcinoma, including keratin expression and typical intercellular bridges. Notably, when inoculated into syngeneic DBA/2 mice (either intravenously or subcutaneously), KLN205 cells demonstrate high tumorigenicity and the capacity for organ-specific metastasis to the lungs, making them a classic immunocompetent animal model for respiratory tumor research. Due to their syngeneic host background, KLN205 cells are widely used to investigate the pathogenesis of lung squamous cell carcinoma, evaluate novel cancer immunotherapies, explore tumor microenvironment interactions, test targeted chemotherapeutic agents, and assess anti-metastatic drug candidates in vivo.

Culture Conditions and Handling
Subculturing 1. Remove and discard the culture medium.
2. Briefly rinse the cell monolayer with PBS (without Ca2+/Mg2+) to completely remove residual serum.
3. Add 1.0 to 2.0 mL of 0.25% trypsin-0.03% EDTA solution and incubate at 37°C until the cell monolayer detaches (typically 2 to 5 minutes).
4. Add fresh complete growth medium to neutralize the trypsin and gently pipette up and down to prepare a single-cell suspension.
5. Seed the cell suspension into new culture vessels.
