JHH-7 Cell Line

General Information
Organism Homo sapiens, human
Cell Line Description JHH-7 is a human hepatocellular carcinoma (HCC) cell line established by Homma, Fujise, and Nagamori in 1990. It was derived from a 53-year-old Japanese male patient with liver cirrhosis and HBsAg-positive hepatocellular carcinoma. JHH-7 cells exhibit an epithelioid, polygonal morphology and grow as an adherent monolayer. Classified as Edmondson grade III, the cell line retains key characteristics of differentiated hepatocytes, including the synthesis of albumin, alpha-fetoprotein (AFP), carcinoembryonic antigen (CEA), and ferritin. Genetically, the cells harbor a p.Arg267Pro point mutation in the TP53 gene, a C228T hotspot mutation in the TERT promoter, and a deletion within the APC-binding domain of the AXIN1 gene. Furthermore, the cells contain only two incomplete, integrated copies of the HBV genome, which are incapable of producing infectious viral particles. JHH-7 serves as an effective model for evaluating p53 activators and inhibitors of the Wnt/β-catenin signaling pathway. It is widely used in research concerning the mechanisms of HBV-associated hepatocarcinogenesis, hepatocyte differentiation, p53 activation, and the response to anticancer drugs in both in vitro and xenograft models.

Culture Conditions and Handling
Subculturing 1. Remove and discard the culture medium.
2. Briefly rinse the cell monolayer with PBS (Ca²⁺/Mg²⁺-free) to thoroughly remove residual serum.
3. Add 2.0 to 3.0 mL of 0.25% Trypsin–0.53 mM EDTA solution, and observe under an inverted microscope until the cell monolayer has dispersed (typically takes 2 to 5 minutes).
4. Add 6.0 to 8.0 mL of complete growth medium, and gently pipette to prepare a single-cell suspension.
5. Aliquot the cell suspension into new culture vessels.
Medium Renewal 1 to 2 times per week
Subcultivation Ratio Split at a ratio of 1:3 to 1:6 when the monolayer reaches approximately 80% confluence.
Culture Conditions Atmosphere: Air, 95%; CO2, 5%; Temperature: 37°C
Cryopreservation Complete growth medium (Williams' E) + 20% FBS + 10% DMSO
