Cell Line DescriptionHuman umbilical vein endothelial cells (HUVECs) are a widely used source of primary endothelial cells for in vitro studies of the vascular system and angiogenesis. They are used as a laboratory model system to study endothelial cell functions and pathologies, such as angiogenesis. HUVECs are relatively easy to isolate, avoid contamination by other cell types, and the umbilical cord is easily available as discarded biological waste after the birth of the child. In addition, the umbilical vein lumen has sufficient capacity, making it easy to insert a cannula, and the endothelium can be isolated by enzymatic digestion. HUVECs have been reported to express many important endothelial markers and signaling molecules related to the regulation of vascular homeostasis. In addition, HUVECs respond to various physiological and pathophysiological stimuli, such as high glucose concentrations, lipopolysaccharide (LPS) administration, or shear stress. HUVECs can also be successfully differentiated into 3D spheroids and co-cultured with other cell types in 3D systems, enabling the establishment of advanced research models to better understand intercellular communication and cell behavior in vivo. HUVECs have been used for the vascularization of various engineered tissues, such as periosteum, bone tissue, and dental tissue, such as biomimetic tooth buds.

Additional InfoCultured HUVEC contains anti-hemophilic factor antigen, now called von Willebrand factor (vWF), which distinguishes endothelial cells from vascular smooth muscle cells and fibroblasts. It is noteworthy that the aforementioned Weibel-Palade bodies are present only in the vascular endothelium and are cytoplasmic components that contain von Willebrand factor.

Culture Conditions and Handling
SubculturingVolumes given are for 75 cm2 flasks. For culture vessels of other sizes, increase or decrease the amount of dissociation medium required proportionally.
1. Remove and discard the medium.
2. Rinse the cell layer briefly with 0.25% (w/v) trypsin-0.53 mM EDTA solution to remove all traces of serum containing trypsin inhibitors.
3. Add 2.0 to 3.0 mL of trypsin-EDTA solution to the flask and observe the cells under an inverted microscope until the cell layer is dispersed (usually within 5 to 15 minutes). Note: To avoid clumping, do not agitate the cells by tapping or shaking the flask while waiting for the cells to detach. Cells that are difficult to detach can be placed at 37°C to facilitate dispersion.
4. Add 6.0 to 8.0 mL of complete growth medium and gently pipette out the cells.
5. Add an appropriate aliquot of the cell suspension to a new culture vessel.
6. Incubate the culture at 37°C.

Subcultivation RatioThe ratio of 1:2 to 1:3 is recommended.
Culture MediumThe basal medium for this cell line is F-12K medium. To make complete growth medium, add the following to 450 mL basal medium:
1. 5 mL 10 mg/mL heparin stock solution, for a final heparin concentration of 0.1 mg/mL in the complete growth medium
2. 50 mL fetal bovine serum
3. 500 μL 30 mg/mL endothelial cell growth supplement
