Customer Reviews

The MDA-MB-436 cell line is a well-established human breast cancer model derived from the pleural effusion of a 43-year-old female patient with metastatic adenocarcinoma. As a triple-negative breast cancer (TNBC) subtype, it lacks the expression of estrogen receptor (ER), progesterone receptor (PR), and human epidermal growth factor receptor 2 (HER2), making it particularly valuable for studying aggressive, hormone-independent cancers. This cell line carries a BRCA1 mutation, making it a key model for studying DNA repair deficiencies and the pathogenesis of hereditary breast cancer. Its molecular characteristics include dysregulation of signaling pathways such as PI3K/AKT and Wnt/β-catenin, which drive cell proliferation and invasion. Researchers frequently use this cell line to study tumor microenvironment interactions, epithelial-mesenchymal transition (EMT), and drug resistance mechanisms, providing insights into novel therapeutic targets for refractory breast cancer.

The luciferase reporter cell line – MDA-MB-436 – is constructed by integrating a bioluminescent reporter system into the parental MDA-MB-436 cells, enabling real-time monitoring of specific molecular events. In drug discovery, it facilitates high-throughput screening of compounds targeting oncogenic signaling pathways (e.g., TGF-β or Notch inhibitors) by measuring luminescence changes. Researchers utilize this model to study in vivo metastasis using bioluminescence imaging, tracking tumor dissemination to organs such as the lungs and bones with high sensitivity. Furthermore, it can be used in functional genomics studies (e.g., CRISPR screening) to identify regulators of cancer stemness or therapeutic resistance. Co-culture studies with immune cells or fibroblasts further elucidate tumor immune evasion and stromal interactions. This reporter gene cell line accelerates the validation of gene therapies and provides a translational bridge between mechanistic studies and preclinical efficacy testing of TNBC interventions.
