OCI-AML3 was established from the peripheral blood of a 57-year-old M4 male. This cell line contains the most common and well-characterized DNMT3A mutation type, the R882 dominant and negative mutation, which results in reduced methyltransferase activity. In addition, OCI-AML3 also contains the nucleolar phosphoprotein (NPM1) gene mutation type A. NPM1 mutations are the most common genetic alterations in adult acute myeloid leukemia (AML) cases, accounting for approximately 30% of total cases, which has led to the use of this cell line. GFP reporter cell line - OCI-AML-3 is a widely used tool in biomedical research, especially in AML research. The introduction of the green fluorescent protein (GFP) reporter gene into these cells has greatly enhanced their utility, providing a powerful means for real-time monitoring of cellular processes and gene expression dynamics. GFP is a naturally fluorescent protein that emits bright green light when exposed to blue to ultraviolet light, making it an ideal marker for observing and tracking biological activities within living cells. Therefore, the GFP reporter cell line - OCI-AML-3 enables researchers to perform live cell imaging to study cellular responses to drugs, gene editing interventions, or environmental changes without the need for invasive techniques or external dyes.
OCI-AML-3 cell line is derived from a patient with acute myeloid leukemia (AML) and has great potential for a variety of research applications. When transfected with a luciferase reporter gene, the OCI-AML-3 cell line becomes a powerful tool for a variety of research and therapeutic purposes: Drug Screening and Development: Using the luciferase reporter OCI-AML-3 cell line, researchers can efficiently screen potential therapeutic compounds. The luminescence signal can be quantitatively measured to evaluate the efficacy of new drugs in inducing cancer cell death or inhibiting proliferation, thereby accelerating the drug discovery process. Gene Regulation Studies: Luciferase reporters can be linked to specific gene promoters to study the regulatory mechanisms of different genes associated with AML. Changes in luminescence can indicate promoter activity in response to various stimuli or genetic modifications, allowing detailed studies of gene function and regulation. Pathway Analysis: By integrating reporter genes with signal transduction pathways, scientists can monitor the activity of specific signaling cascades within OCI-AML-3 cells. This is particularly useful for understanding how specific pathways lead to leukemogenesis and identifying potential targets for treatment. In Vivo Imaging: When transplanted into animal models, luciferase-expressing OCI-AML-3 cells allow for in vivo imaging of tumor growth and metastasis. Luminescence can be detected non-invasively, providing real-time insights into AML progression and the effects of therapeutic interventions. High-Throughput Screening: The Luciferase Reporter Cell Line - OCI-AML-3 is amenable to high-throughput screening formats, making it suitable for large-scale studies aimed at identifying new drugs, genetic targets, or mechanistic insights into AML. Drug Resistance Studies: The Luciferase Reporter Cell Line - OCI-AML-3 can be used to explore drug resistance mechanisms in AML. By monitoring changes in luminescence following drug treatment, researchers can identify resistance pathways and work to overcome treatment resistance.
