Revival Rapidly thaw cells in a 37°C water bath. Transfer contents into a tube containing pre-warmed media. Centrifuge cells and seed into a 25 cm2 flask containing pre-warmed media.

Safety Considerations The following safety precautions should be observed.
1. Use pipette aids to prevent ingestion and keep aerosols down to a minimum.
2. No eating, drinking or smoking while handling the stable line.
3. Wash hands after handling the stable line and before leaving the lab.
4. Decontaminate work surface with disinfectant or 70% ethanol before and after working with stable cells.
5. All waste should be considered hazardous.
6. Dispose of all liquid waste after each experiment and treat with bleach.

BAF3 is a mouse pre-B lymphocyte cell line originally derived from mouse bone marrow. It is widely used as a hematopoietic model system to study cytokine-dependent proliferation, signal transduction, oncogenic kinase activity, and immune-related cellular responses. The survival and growth of parental BAF3 cell lines depend on interleukin-3 (IL-3), making it a valuable platform for assessing factors or genetic alterations that promote cytokine-independent proliferation or activate downstream signaling pathways. The BAF3 luciferase reporter gene cell line has been genetically engineered to stably express the luciferase reporter gene in the BAF3 cell background. Through stable integration and selection, this reporter gene cell line provides consistent luciferase expression throughout passage, enabling sensitive, quantitative, and reproducible measurements of cell number, viability, proliferation, transplantation, or treatment response based on luminescence signal output.

The BAF3 luciferase reporter gene cell line has broad applications in biomedicine, immunology, oncology, and drug development. One major application is cell proliferation and viability analysis, where luminescence intensity serves as an indirect but highly sensitive reading of viable cell count. This is particularly useful for screening cytokines, growth factors, antibodies, small molecules, kinase inhibitors, or biologics that affect BAF3 cell survival or expansion. In cancer biology, BAF3-derived models are commonly used to study oncogenic tyrosine kinases, receptor fusions, mutant signaling proteins, and pathway activation associated with hematologic malignancies.
