Reverse Transcriptase

Reverse transcriptase (RT) is an enzyme used to generate complementary DNA (cDNA) from an RNA template, a process known as reverse transcription. The reverse transcriptase (RT) enzyme found in retroviruses first converts the viral RNA genome into complementary DNA (cDNA) and then into double-stranded DNA that integrates into the host genome. Retroviral RTs possess three sequential biochemical activities: RNA-dependent DNA polymerase activity, ribonuclease H (RNase H) and DNA-dependent DNA polymerase activity. Collectively, these activities enable the enzyme to convert single-stranded RNA to double-stranded cDNA. In retroviruses and retrotransposons, this cDNA can integrate into the host genome, from which new RNA copies can be made via host-cell transcription.

The ability of reverse transcriptase to synthesize DNA from RNA has already been used in the laboratory. Reverse transcription combined with PCR is a popular technique in molecular biology (termed RT-PCR) and has been widely used to detect expressed genes, examine transcript variation, and generate cDNA templates for cloning and sequencing. One of the first applications of RT in molecular biology was the construction of cDNA libraries for the characterization of novel RNA transcripts, gene sequencing, and expression of recombinant proteins after cloning. Furthermore, reverse transcription combined with PCR or RT-PCR allows the detection of RNA even at very low levels of gene expression and paves the way for the detection of circulating RNA, RNA viruses, and cancerous gene fusions in molecular diagnostics.
