3D4/21 is a porcine alveolar macrophage cell line widely used as an in vitro model for studying porcine innate immunity, respiratory infections, macrophage biology, and host-pathogen interactions. Parental 3D4/21 cells retain many macrophage-like characteristics, including adhesive growth, responsiveness to inflammatory stimuli, and the ability to participate in porcine-associated immune signaling pathways. The luciferase reporter gene cell line – 3D4/21 – is an engineered derivative of this macrophage cell line, stably incorporating the luciferase reporter gene system. Depending on the reporter gene design, the luciferase gene can be placed under the control of a constitutive promoter or a pathway-responsive promoter, allowing for sensitive monitoring of cell activity, promoter activation, signaling pathway induction, or infection-related biological changes via fluorescence readings.
