The RAW 264.7 cell line is a highly utilized murine macrophage model, originally established from an ascites tumor induced in a male BALB/c mouse by the Abelson murine leukemia virus. These cells exhibit a characteristic monocyte-macrophage morphology, readily adhering to culture surfaces while demonstrating active phagocytotic and pinocytotic capabilities. They also express essential pathogen recognition receptors, making them highly responsive to microbial ligands. The Luciferase Reporter Cell Line - RAW 264.7 is an advanced derivative engineered through stable lentiviral transduction to constitutively express the firefly luciferase enzyme. This stable genetic integration ensures that the modified cells continuously emit a robust, quantifiable bioluminescent signal upon exposure to the substrate D-luciferin. Importantly, this targeted modification creates a highly sensitive cellular tracking tool without compromising the functional immunological phenotype, receptor profile, or baseline survival characteristics of the original parental line.

Because they accurately mirror primary macrophage behavior, these luciferase-expressing cells are indispensable tools for both pre-clinical immunology and pharmacological research. In vivo, the constitutive bioluminescent signal enables the use of non-invasive imaging to continuously track macrophage migration, proliferation, and survival over time. This approach is highly valuable in whole-animal models of localized inflammation, tumor microenvironments, or infectious diseases, allowing scientists to dynamically visualize immune cell homing and recruitment while minimizing the need for multiple endpoint sacrifices. In vitro, the reporter model streamlines high-throughput cell viability and cytotoxicity screening. Researchers rely on this quantifiable bioluminescent readout to rapidly assess the impact of novel immunomodulatory drugs, anti-inflammatory compounds, and targeted nanotherapies, ultimately accelerating the discovery of treatments designed to modulate innate immune responses.

Ideal for Inflammation Assays

Stable transfection in RAW 264.7 cells can be difficult, so buying this ready-to-use reporter line was a great decision. The cells respond beautifully to LPS stimulation, and the luciferase output provides a very clear readout for our anti-inflammatory drug screens.
