The SACC-LM cell line is a highly metastatic human salivary adenoid cystic carcinoma (SACC) model, originally established as a lung-metastatic subclone isolated from the parental SACC-83 line. Growing as an adherent epithelial monolayer, these cells are particularly characterized by their aggressive malignant behavior and a strong inherent propensity for pulmonary metastasis. The Luciferase Reporter Cell Line - SACC-LM represents an advanced derivative engineered through stable lentiviral transduction to constitutively express the firefly luciferase enzyme. This targeted genetic integration ensures that the modified cells continuously emit a robust, quantifiable bioluminescent signal upon exposure to the substrate D-luciferin. Crucially, this stable modification transforms the standard line into a highly sensitive cellular tracking tool while strictly preserving the aggressive malignant phenotype, cellular morphology, and specific lung tropism of the original SACC-LM line.

Because of its exceptional metastatic capacity, this reporter cell line is heavily utilized in pre-clinical oncological research, particularly for modeling advanced head and neck cancers. In vivo, the cells are widely used to establish highly reliable orthotopic and systemic xenograft mouse models. By utilizing non-invasive bioluminescence imaging (BLI), researchers can continuously visualize primary salivary tumor growth and dynamically track the spatiotemporal progression of distant lung metastases in living animals over time. This longitudinal approach eliminates the need for repeated endpoint sacrifices while providing precise, real-time disease mapping. In vitro, the constitutive luminescence facilitates rapid high-throughput screening for cell viability and proliferation. Researchers depend on this quantifiable optical readout to evaluate the cytotoxicity of novel chemotherapeutics, anti-metastatic agents, and targeted molecular inhibitors, accelerating the development of effective interventions for late-stage, metastatic salivary gland carcinomas.
