A reporter cell line for luciferase Luciferase is a bioluminescent enzyme that is extensively utilized in molecular biology research, and Jurkat is designed to express it. Typically, the luciferase gene is incorporated into the genome of Jurkat cells, enabling real-time monitoring of signaling pathway activity or gene expression levels by luminescence assays. A patient with T-cell leukemia provided the well-known human T lymphocyte cell line known as Jurkat cells. They function as mature T cells and are frequently employed in studies on cancer and immunity. A useful resource for researching a variety of biological processes, such as drug screening, signal transduction, and gene regulation, are luciferase reporter cell lines. Through the connection of luciferase expression to particular promoter regions or signaling pathways, scientists may examine how different stimuli or treatments affect the activation of pathways or gene expression in Jurkat cells. High-throughput screening experiments made possible by these cell lines also aid in the search for new therapeutic agents and the clarification of the molecular pathways that underlie the development of disease.

The scientists showed that combining luciferase with the carbohydrate-binding protein galectin-3 (G3) greatly increased the amount of bioluminescence in subcutaneous tissue. While G3-luciferase fusions lasted for about seven days, unmodified luciferase was only visible for a few hours. Trimeric G3-luciferase assemblies were created by additional engineering, extending bioluminescence to 3 days intramuscularly and 14 days subcutaneously. The extended impact was ascribed to the trimeric structure's increased affinity for binding carbohydrates. Notably, these alterations did not affect wild-type G3 signaling and did not result in systemic accumulation after injection, indicating a secure and practical method to improve the local pharmacokinetics of enzyme medications.

The Figure 1 describes various experimental observations related to Jurkat T cells treated with different forms of G3 fusion proteins. (doi: 10.1038/s41467-018-07129-6)Figure 1. Using fluorescence filters, the researchers show how Luciferase Reporter Cell Line - Jurkat can evaluate extracellular signaling using NL-G3 and NL-TT-G3 bioluminescence as well as GFP-G3 and GFP-TT-G3 fluorescence on Jurkat T cells. 1. Signaling Pathway Monitoring: The luciferase reporter cell line, Jurkat, which is designed to express luciferase, helps in molecular biology research by allowing real-time monitoring of the activity of signaling pathways or gene expression levels through luminescence assays. 2. Biological Process Research: Jurkat cells are an invaluable tool for researching a range of biological processes, including gene regulation, drug screening, and signal transduction. The investigation of the effects of various stimuli or treatments on pathway activation or gene expression in Jurkat cells is made easier by the use of luciferase reporter cell lines. 3. Tumor and Immunity Studies: Developed from a T-cell leukemia patient, Jurkat cells are a mature form of T cells that are widely employed in immunology and cancer research to shed light on disease mechanisms and possible targets for treatment.
Customer Q&As
Luciferase Reporter Cell Line - What is the Luciferase expression level of the Jurkat cell line? Is it stable under different culture conditions?

A: Our Luciferase Reporter Cell Line - Jurkat has undergone rigorous screening and verification to ensure stable expression of the Luciferase gene. Through methods such as luciferase activity detection and PCR, we verified the expression level of Luciferase and performed repeated tests at different time points and in different batches. Cells can maintain stable Luciferase expression under appropriate culture conditions and are suitable for long-term experimental use.

How does the Jurkat background impact the performance of this Luciferase cell line? Are there any special training requirements?

A: As a human T-cell leukemia cell line, the background of the Jurkat cell line may affect the signaling and response characteristics of the cells, but it will not significantly affect the expression of Luciferase. We recommend that customers pay attention to the special growth requirements of Jurkat cell lines during use, such as culture medium components and culture conditions. Detailed culture guidelines will help customers maintain their cells in optimal condition.

Have the cell lines for this product been tested for contaminants? How to ensure the purity of cell lines?

A: Our Luciferase Reporter Cell Line - Jurkat undergoes rigorous contaminant testing, including testing for common contaminants such as mycoplasma and bacteria. We use methods such as PCR and cell culture testing to ensure the purity of our cell lines. We have strict quality control procedures to ensure that each batch of cell lines meets high standards of purity.
