What is qPCR?
Real-time PCR or quantitative PCR (qPCR) is one of the most reliable methods of detection and quantification of specific DNA and RNA molecules. It is a more precise and accurate method of PCR because it can detect small changes in gene expression levels in a sample. It is widely used in various applications such as gene expression profiling, viral load quantification, microarray data validation, pathogen detection, and forensic analysis.
Dye-based qPCR
Dye-based quantitative PCR (qPCR) uses real-time fluorescence of double-stranded DNA (dsDNA)-binding dyes to measure DNA amplification that occurs during PCR. The dye exhibits weak background fluorescence that increases dramatically upon binding to dsDNA. Amplification of the target sequence thus results in an increase in fluorescence that is proportional to the amount of dsDNA present in each PCR cycle. This type of qPCR analysis requires only two sequence-specific primers, making it a fast and cost-effective method to interrogate large numbers of samples/targets.
Dye-based qPCR kits come in different formats, including kits designed for one- or two-step RT-qPCR. The One-Step RT-qPCR Kit enables reverse transcription of RNA to cDNA and qPCR amplification in a single reaction chamber. On the other hand, two-step RT-qPCR kits allow reverse transcription of RNA to cDNA in a separate reaction prior to qPCR amplification.
