HK-1 is a human nasopharyngeal carcinoma cell line that has been widely utilized as an in vitro model to investigate the biological characteristics of nasopharyngeal epithelial malignancies—including tumor cell proliferation, survival, migration, invasion, drug response, and the pathogenesis of virus-associated cancers. The HK-1 Luciferase Reporter Cell Line was constructed by genetically engineering HK-1 cells to stably express a luciferase reporter gene. Since the luciferase gene has been integrated into the cellular genome and maintained under stable selection conditions, this reporter cell line yields consistent and reproducible signal outputs during passaging under recommended culture conditions. Its luminescence intensity is typically directly proportional to the number of viable reporter cells, making this cell line a convenient tool for quantitative cellular assays. Compared to traditional endpoint detection methods—such as those based on staining, microscopic imaging, or colorimetric readouts—the HK-1-Luc reporter cell line offers a rapid, sensitive, and non-destructive (or minimally destructive) approach for real-time monitoring of cellular growth and viability.
The HK-1 Luciferase Reporter Cell Line finds broad application across numerous fields within cancer biology, pharmacology, and translational medicine research. One of its most common applications is in assays for cell proliferation and cytotoxicity. In such assays, changes in luminescence signals are monitored to assess the impact of small-molecule compounds, biologics, natural products, gene knockdown, gene overexpression, or other experimental interventions on the viability and growth status of HK-1 cells. Given the exceptional sensitivity of luciferase-based detection methods and their high compatibility with multi-well plate formats, this cell line is ideally suited for medium-to-high-throughput drug screening and dose-response analyses. Furthermore, this cell line can be utilized to evaluate anti-tumor activity within co-culture systems—such as in immune cell-mediated killing assays, studies on antibody-dependent cell-mediated cytotoxicity (ADCC), or exploratory investigations into the mechanisms of interaction between tumor cells and stromal cells.
