BJ cells are a line of normal, mature human fibroblasts originally derived from neonatal foreskin tissue; they are widely utilized in biomedical research as a representative model of non-transformed human fibroblasts. Because BJ cells retain many characteristics of primary human fibroblasts, they are frequently selected for studies involving cell growth, senescence, extracellular matrix biology, wound healing, stress responses, and host-cell interactions. The BJ luciferase reporter cell line (BJ-Luc) is an engineered derivative of the BJ cell line, modified to stably express a luciferase reporter gene. Through the stable integration or long-term expression of the reporter construct, this cell line generates a stable luminescent signal upon the addition of the appropriate luciferase substrate. Compared to transiently transfected cells, the stable BJ-Luc reporter cell line offers superior reproducibility and reduced experimental variability, while also facilitating more convenient execution of repetitive or large-scale assays.

The BJ luciferase reporter cell line is suitable for a variety of cell-based assays and preclinical research workflows. A primary application is high-throughput screening, where the high sensitivity and broad dynamic range of the luciferase signal enable the rapid, quantitative assessment of compounds, biologics, environmental factors, or genetic perturbations. By correlating the luminescent signal with viable cell numbers or reporter gene expression levels, the cell line can be used to evaluate cytotoxicity, cell proliferation, survival, and metabolic effects. Given that BJ cells are normal human fibroblasts, this reporter cell line is highly valuable for research into skin biology, connective tissue remodeling, wound repair, and mechanisms of senescence, as well as for studying fibroblast responses to stimuli such as cytokines, drugs, nanoparticles, radiation, or changes in the extracellular matrix. In the fields of drug discovery and safety assessment, BJ-Luc cells facilitate comparative studies between normal and disease-derived cells, aiding in the identification of compounds that exhibit selective activity against normal human stromal cells or demonstrate adverse toxicity.
