Safety Considerations The following safety precautions should be observed.
1. Use pipette aids to prevent ingestion and keep aerosols down to a minimum.
2. No eating, drinking or smoking while handling the stable line.
3. Wash hands after handling the stable line and before leaving the lab.
4. Decontaminate work surface with disinfectant or 70% ethanol before and after working with stable cells.
5. All waste should be considered hazardous.
6. Dispose of all liquid waste after each experiment and treat with bleach.

BV2 is a widely used murine microglia cell line that serves as an in vitro model for studying the immune cell biology of the central nervous system. BV2 cells retain many key characteristics of primary microglia, including rapid proliferation, a strong response to inflammatory stimuli, phagocytic activity, production of cytokines and chemokines, and activation of signaling pathways associated with neuroinflammation. Due to the difficulty in isolating large quantities of primary microglia and the significant batch-to-batch variability, BV2 cells provide a convenient, reproducible, and cost-effective alternative for mechanistic and screening studies. The luciferase reporter gene cell line – BV2 is an engineered BV2-derived cell model in which the luciferase reporter gene has been stably introduced into the cell genome or maintained through a stable expression system.

The luciferase reporter gene cell line-BV2 is particularly suitable for research in areas such as neuroinflammation, innate immunity, mechanisms of neurodegenerative diseases, and drug development. This method can be used to evaluate how candidate compounds, biologics, natural products, nanoparticles, toxins, or gene interventions affect microglia activation and downstream signaling. For example, researchers can stimulate BV2 reporter cells with lipopolysaccharides, cytokines, amyloid-related peptide, oxidative stress inducers, or other inflammatory triggers, and then monitor the resulting luciferase activity to assess the activation or inhibition of specific pathways. This makes the cell line suitable for screening anti-inflammatory or neuroprotective agents that may regulate microglial responses associated with Alzheimer's disease, Parkinson's disease, multiple sclerosis, traumatic brain injury, ischemic injury, and other central nervous system disorders. In high-throughput screening pipelines, luciferase-based BV2 cells can shorten detection time and improve data consistency because their bioluminescent signals are easily quantified and can be measured directly in microplates.
