Case Study 1
Caspase-8 has unique roles in inducing and suppressing apoptosis and necroptosis mediated by death receptors. Researchers aimed to investigate the role of Caspase-8 in regulating RA-induced cell differentiation and necroptosis by utilizing cDNA Clone knockdown. They demonstrated that Caspase-8 knockdown in embryoid bodies derived from ES cells enhanced both RA-induced cell differentiation and necroptosis, dependent on Ripk1 and Ripk3. Notably, the enhancement of RA-induced cell differentiation was independent of Mlkl and necrosome formation. By inducing the expression of RA-specific target genes and RIPK1, RIPK3, and MLKL, RA treatment activated necroptosis. Furthermore, Caspase-8 knockdown led to the translocation of RIPK1 and RIPK3 into the nucleus, forming a complex with RA receptor (RAR) and enhancing RAR binding to RARE. In Caspase-8-deficient embryos, as well as embryos deficient in both Caspase-8 and Mlkl, the expression of RA-specific target genes was significantly enhanced.

Proper flagellar shape and movement in sperm are crucial for successful fertilization.
