The poly (ADP-ribose) polymerase (PARP) is an emerging family of enzyme that has the ability to catalyze the transfer of ADP-ribose to target proteins. PARP family consists of at least 18 members of PARP proteins that are encoded by different genes, but share homology in a conserved catalytic domain. PARPs play an important role in various cellular processes, including modulation of chromatin structure, transcription, replication, recombination, and DNA repair. Among these functions, the role of PARP proteins in DNA repair is of particular interest. It was found that certain tumors defective in homologous recombination mechanisms may rely on PARP-mediated DNA repair for survival, which means that PARP can be a potential target of cancer therapy.

Several latter findings have shown that inhibition of PARP, either alone or in combination with DNA-damaging agents, could be a potential therapeutic approach in TNBC and other tumor types. Inhibition of PARP has potential for use in cancer treatment through at least two mechanisms, i.e., by increasing tumor sensitivity to chemotherapeutic agents that damage DNA, and also by inducing “synthetic lethality” in cells that are highly dependent on PARP, due to deficiency in HR, such as BRCA1 mutants. Besides, the hyperactivation of PARP may also be exploited to selectively kill cancer cells as well. Other PARP forms, including tankyrase 1 (PARP 5a), which plays an important role in enhancing telomere elongation by telomerase, have been found to be potential targets in cancer therapy as well. PARPs are becoming popular target of different cancer treatment.
