Yeast three-hybrid system is a derivative of yeast two-hybrid (Y2H) and it is a kind of powerful tool to detect RNA-protein interactions. In this system, a transcription factor is assembled by the use of an RNA bridge that brings a fusion protein containing a DNA-binding domain together with a fusion protein containing an activation domain. The assembly of this ternary complex usually depends on each fusion protein containing an RNA-binding domain that binds to a site in the RNA molecule. When this ternary complex forms at a promoter, even transiently, the expression of the reporter gene is turned on. Reporter expression can be detected by cell growth, colony color, or the levels of a specific enzyme. Thus, the RNA-protein interactions can be monitored by phenotype or simple biochemical assays.
