Revival Rapidly thaw cells in a 37°C water bath. Transfer contents into a tube containing pre-warmed media. Centrifuge cells and seed into a 25 cm2 flask containing pre-warmed media.

Safety Considerations The following safety precautions should be observed.
1. Use pipette aids to prevent ingestion and keep aerosols down to a minimum.

4. Decontaminate work surface with disinfectant or 70% ethanol before and after working with stable cells.
5. All waste should be considered hazardous.

MG63 is an adherent human osteosarcoma cell line established from the bone tumor of a 14-year-old male patient. The cells exhibit fibroblast-like morphology and are frequently used as an osteoblast-like model in studies of bone tumor biology, extracellular matrix responses, and osteogenic signaling. MG63 cells express transforming growth factor beta receptors and can respond to factors involved in bone formation and remodeling. Luciferase Reporter Cell Line - MG63 is an engineered derivative that expresses a luciferase reporter gene. Following addition of a compatible luciferin substrate, the cells generate a bioluminescent signal that can be measured with a luminometer or imaging system. This reporter format provides a sensitive and scalable alternative to assays that rely entirely on staining, microscopy, or manual cell counting.

The cell line is useful for evaluating osteosarcoma cell proliferation, survival, treatment response, and relative cell abundance. Luciferase activity can provide a convenient readout for dose–response studies involving conventional chemotherapeutics, targeted compounds, biologics, or combination treatments. Because parental MG63 cells are also widely used in biomaterials research, the reporter derivative may support investigations of cell attachment, growth, and cytotoxicity on bone substitutes, implant coatings, scaffolds, and tissue-engineering materials. In three-dimensional cultures, luminescence can assist with monitoring viable cells when direct microscopic assessment is limited by scaffold thickness or model complexity. In appropriately validated xenograft studies, bioluminescence imaging may enable repeated assessment of tumor establishment, progression, and therapeutic response in the same animal.
