Caco-2 cells are a well-characterized human epithelial cell line originally isolated from a colorectal adenocarcinoma; they are now widely utilized as an in vitro model for studying intestinal epithelium. Under appropriate culture conditions, Caco-2 cells undergo differentiation to form a polarized monolayer, exhibiting biological characteristics akin to native intestinal epithelial cells—including the formation of tight junctions, the expression of brush-border enzymes, and directional transport functions. These attributes render this cell line invaluable for research across various fields, including intestinal barrier function, drug absorption, transporter protein activity, and epithelial cell biology. The Caco-2 Luciferase Reporter Cell Line is a genetically engineered reporter system constructed by stably integrating a luciferase reporter gene into the genomic background of the parental Caco-2 cell line. Thanks to this stable genomic integration, the cells generate a quantifiable bioluminescent signal upon the addition of an appropriate luciferase substrate. Compared to transient transfection systems, this stable Caco-2-Luc reporter cell line offers superior experimental reproducibility, reduced data variability, and a simpler experimental workflow.
The Caco-2 Luciferase Reporter Cell Line finds broad application across numerous disciplines, including biomedicine, drug discovery and development, toxicology, and cell biology. In the context of drug discovery and development, this cell line is particularly well-suited for assessing the impact of various compounds on intestinal epithelial cells—specifically their cytotoxicity, growth-inhibitory effects, proliferation dynamics, and cellular responses to candidate drugs. Given that luciferase activity typically correlates positively with either the number of viable cells or the activity of the specific signaling pathway monitored by the reporter gene, researchers can utilize this cell line to rapidly and quantitatively analyze cellular responses to various treatments in a highly sensitive and scalable manner. Furthermore, in studies investigating intestinal drug absorption and intestinal barrier function, Caco-2-Luc cells serve as an ideal experimental model for examining how factors such as drugs, nutrients, toxins, inflammatory mediators, or microbial metabolites influence the integrity and function of intestinal epithelial cells.
