What Is Probe Based-qPCR?

Quantitative PCR (qPCR) is a powerful and commonly used technique for the precise analysis of gene expression. Probe-based qPCR is a variant of the core qPCR technique that employs fluorescent probes to enable specific target detection during PCR amplification. A fluorescent probe is a short piece of DNA labeled with a fluorophore and a quencher. Initially, the quencher's proximity to the fluorophore prevents it from emitting fluorescence despite being excited by the light source. However, during amplification, the probe binds to the target sequence, is cleaved by the polymerase, and in the process separates the fluorophore from the quencher. This separation produces a fluorescent emission that correlates directly to the amplification of the target DNA.

The Applications of Probe Based-qPCR

Probe-based qPCR is commonly used in clinical and diagnostic applications, as it is well suited for any application requiring maximum specificity and consistent performance. Probe-based qPCR is able to achieve this level of performance because the probes are highly specific for the target sequence and the formation of primer-dimers is very rare. Since each probe is uniquely labeled, multiple targets can be amplified in the same tube by multiplexing. This increases throughput, minimizes sample handling, and potentially reduces reagent and consumable usage.
