Nuclease

What Is Nuclease?

Nucleases are specialized enzymes that cleave the phosphodiester bonds between nucleotides in nucleic acids (DNA and RNA). Nuclease activity is integral to DNA replication; 5' to 3' exonuclease and endonuclease are required for RNA primer removal and 3' to 5' exonuclease for proofreading. Two other major DNA metabolic processes, recombination and repair, are initiated by nucleases. Nuclease activities are also required for structural changes in nucleic acids, such as topoisomerization, site-specific recombination, and RNA splicing, in which phosphodiester bonds are temporarily broken and reformed after strand passage or transfer to new targets. Additionally, nuclease activity is critical for RNA processing, maturation, and RNA interference.

Types of Nucleases

According to the active site, nucleases can be divided into two main groups: exonucleases and endonucleases. Exonucleases digest nucleic acids from the ends, while endonucleases act on the central region of the target molecule. Exonucleases remove one nucleotide at a time from a DNA molecule, while endonucleases work by cleaving phosphodiester bonds within the DNA molecule. These classes can be further subdivided into deoxyribonucleases and ribonucleases. Deoxyribonucleases (DNA nucleases) act on DNA, while ribonucleases act on RNA.
