| Safety Considerations |
The following safety precautions should be observed. 1. Use pipette aids to prevent ingestion and keep aerosols down to a minimum. 2. No eating, drinking or smoking while handling the stable line. 3. Wash hands after handling the stable line and before leaving the lab. 4. Decontaminate work surface with disinfectant or 70% ethanol before and after working with stable cells. 5. All waste should be considered hazardous. 6. Dispose of all liquid waste after each experiment and treat with bleach. |

A-431 is a mature human epidermal-like cancer cell line originally derived from the skin/epidermis of an 85-year-old female patient. Due to its favorable epithelial morphology, adherent growth characteristics, and high expression of epidermal growth factor receptor (EGFR), it is widely used in cancer biology, dermatology research, signal transduction research, and drug development. These characteristics make A-431 cells uniquely valuable in studying EGFR-mediated signaling pathways, tumor cell proliferation, receptor activation, and responses to targeted therapies. The luciferase reporter gene cell line-A-431 is an engineered derivative of the parental A-431 cell line that stably expresses the luciferase reporter gene system.
The luciferase reporter gene cell line-A-431 can be used in a variety of experimental and screening procedures. In oncology research, this cell line can be used to evaluate tumor cell growth, pathway regulation, and the effects of anticancer compounds, particularly suitable for drugs targeting EGFR-related signaling pathways, MAPK/ERK activation, PI3K/AKT pathways, inflammatory responses, or transcriptional regulation, depending on the reporter gene construct. Because luminescence intensity can reflect reporter gene pathway activity or relative cellular abundance, researchers can use this cell line to compare the potency of candidate drugs, antibodies, small molecule inhibitors, biologics, natural products, or gene regulation agents.
