Exploring Virus–Host Interactions Through Combined Proteomic Approaches Identifies BANF1 as a New Essential Factor for African Swine Fever Virus

Paper 2025
Molecular & Cellular Proteomics | DOI
Background
African swine fever virus (ASFV) causes deadly disease in pigs and threatens global pork production. This study combined affinity purification–mass spectrometry and yeast two-hybrid screening to investigate ASFV proteins MGF360-21R and A151R, identifying host protein BANF1 as a common and critical interactor. Silencing BANF1 significantly reduced ASFV replication, indicating a proviral role. Furthermore, both viral proteins suppressed type I interferon induction, while recombinant viruses lacking these genes induced stronger IFN responses in porcine macrophages. These findings illuminate ASFV host manipulation and highlight BANF1 as a potential antiviral target.
Key Findings
Using AP-MS and Y2H, 232 and 33 host interactors were identified for MGF360-21R and A151R, respectively, with 48 shared partners including BANF1. GST pulldown and colocalization confirmed interactions, and MST determined a Kd of 105 nM for A151R-BANF1. Expression of either viral protein caused relocalization of nuclear BANF1 to cytoplasm. BANF1 knockdown in WSL cells reduced ASFV titers by approximately 2 logs and altered expression of proteins in innate immune and chromatin-remodeling pathways. Luciferase assays showed that A151R and MGF360-21R inhibit IFN-β induction by cGAS/STING, poly(dA:dT), or RIG-I without affecting ISRE. Deletion mutants of either gene in ASFV Georgia/07 triggered higher IFN-α and CXCL10 secretion in infected macrophages than wild-type, and ΔA151R exhibited a replication defect. These results uncover new immune evasion functions and establish BANF1 as a necessary host factor.

Creative Biogene provided the porcine cDNA library used in yeast two-hybrid screens.

In a study of host dependency factors for multiple SARS-CoV-2 variants, researchers found conserved transcriptional responses in infected Calu-3 cells and, using a genome-wide CRISPR loss-of-function screen, identified host genes required for infection, many shared across variants; validation with small molecules and approved drugs highlighted reactive oxygen species production as an early step in viral replication. Creative Biogene's cDNA library construction platform can support similar studies by providing full-length, normalized, and differentially enriched cDNA libraries suitable for capturing transcriptomic changes and enabling high-confidence discovery of genes involved in variant-independent cellular responses.

FAQ

What materials are needed to start a cDNA library project?

  • Sample: Fresh-frozen tissue (>100 mg), cells (>10^7), or high-quality total RNA (>200 ng/µL, RIN >8.0, intact). Total RNA is preferred.
  • Project Info: Experimental goal, library type, downstream applications (e.g., yeast hybrid, phage display), and any specific vector.
  • Our scientific advisors provide guidance on sample preparation and design if uncertain.

Typical construction timeline for different library types:

The typical construction timeline depends on the library type: standard cDNA libraries generally take about four to five weeks, full-length, normalized, subtracted, or SSH libraries require approximately six to eight weeks, and yeast hybrid libraries usually take five to seven weeks. Timelines may vary based on sample quality and specific library requirements, and detailed schedules are provided at the beginning of each project.

Are downstream screening or sequencing services provided?

Constructed cDNA libraries are ready for downstream applications, including high-throughput sequencing using Illumina or PacBio platforms, functional screening such as yeast one-, two-, and three-hybrid assays, or phage display selection, and targeted gene cloning directly from the library.
