EA.hy926 is a widely used human endothelial-like cell line generated by fusion of human umbilical vein endothelial cells with the thioguanine-resistant A549 lung carcinoma cell line. It retains many characteristic properties of vascular endothelial cells, including an adherent cobblestone-like morphology, expression of endothelial-associated markers, and responsiveness to inflammatory, angiogenic, and vascular regulatory stimuli. Because of its stable growth, convenient handling, and reproducible biological responses, EA.hy926 has become a practical in vitro model for studying endothelial cell function, vascular inflammation, permeability regulation, oxidative stress, coagulation-related pathways, and angiogenesis-associated mechanisms. The Luciferase Reporter Cell Line - EA.hy926 is an engineered derivative of the EA.hy926 cell line that stably expresses a luciferase reporter gene. By integrating the luciferase expression cassette into the cellular system, this reporter cell line enables sensitive, quantitative, and rapid detection of cellular signals through luminescence measurement.

The EA.hy926 luciferase reporter cell line is broadly applicable to a wide range of cell-based assays, spanning fields such as endothelial biology, drug discovery, vascular disease research, and molecular pathway analysis. It is particularly well-suited for assessing cellular responses to cytokines, growth factors, simulated hypoxic environments, oxidative stress inducers, inflammatory mediators, microbial components, or candidate therapeutic agents. Given the pivotal role endothelial cells play in maintaining vascular homeostasis and in the pathogenesis of disease, this reporter cell line serves as a powerful tool for investigating endothelial cell activation, angiogenic signaling, vascular inflammation, vascular barrier dysfunction, and the cellular effects of pro- or anti-inflammatory drugs. In drug screening initiatives, EA.hy926-Luc cells can be utilized in high- or medium-throughput formats to evaluate the activity, potency, potential toxicity, or pathway-modulatory effects of various compounds. Leveraging its stable luciferase reporter signal, researchers can perform quantitative comparisons across different treatment groups, generate dose-response curves, and identify compounds that—under specific experimental conditions—either enhance or inhibit reporter gene activity.
