The PC-3M cell line is a highly aggressive human prostate cancer model, originally established as a metastatic subline from the classic PC-3 parental line. Isolated from liver metastases following the injection of PC-3 cells into nude mice, this adherent, epithelial-like line is characterized by its robust metastatic potential, androgen independence, and lack of androgen receptor (AR) expression. Consequently, it serves as an excellent representation of advanced, castration-resistant prostate cancer. The Luciferase Reporter Cell Line - PC-3M is an advanced derivative engineered through stable lentiviral transduction to constitutively express the firefly luciferase enzyme. This targeted genetic integration ensures that the cells continuously emit a strong bioluminescent signal upon exposure to the substrate D-luciferin. Crucially, this stable modification transforms the cells into a highly sensitive, quantifiable tracking tool while strictly preserving the aggressive, highly metastatic phenotype of the original PC-3M line.

Due to its exceptional metastatic capacity, this reporter cell line is heavily utilized in pre-clinical oncological research, particularly for modeling advanced prostate cancer. In vivo, the cells are widely used to establish highly reliable orthotopic and systemic xenograft mouse models. By utilizing non-invasive bioluminescence imaging (BLI), researchers can continuously visualize primary tumor growth and dynamically track the spatiotemporal progression of distant metastases—especially to bone and visceral organs—in living animals over time. This longitudinal approach eliminates the need for repeated endpoint sacrifices while providing precise disease mapping. In vitro, the constitutive luminescence facilitates rapid high-throughput screening for cell viability and proliferation. Researchers depend on this robust model to evaluate the cytotoxicity of novel chemotherapeutics, gene therapies, and targeted molecular inhibitors, accelerating the development of effective interventions for late-stage, hormone-refractory prostate carcinomas.
