miRNA cDNA synthesis is the process of converting small RNA molecules called miRNA (microRNA) into cDNA (complementary DNA). This is done using reverse transcription, in which an enzyme called reverse transcriptase is used to convert the miRNA to its complementary DNA sequence. This cDNA can then be amplified and analyzed using various molecular biology techniques, providing insight into the function of the miRNA and its role in gene regulation.
miRNA cDNA synthesis involves the use of miRNA-specific reverse transcription primers designed to anneal to the 3' end of the mature miRNA sequence. miRNA-specific primers are often used in combination with poly(T) primers to increase the efficiency of reverse transcription of miRNA templates. The use of specific primers significantly increases the sensitivity and specificity of miRNA detection compared to standard cDNA synthesis.
Creative Biogene's miRNA cDNA Synthesis Kit is an all-in-one, ready-to-use product for the reverse transcription of miRNA from total RNA preparations or enriched miRNA preparations. miRNAs are not polyadenylated in nature. Using our miRNA cDNA synthesis kit, miRNAs are polyadenylated in a poly(A) polymerase reaction. Poly(A)-tailed miRNAs are then converted to cDNA using oligo-dT adapter primers with the addition of reverse transcriptase and other reagents required for cDNA synthesis. Adapter primers have a unique sequence at their 5' end that allows amplification of cDNA in real-time qRT-PCR reactions.
