Gene transfer vectors derived from adeno-associated viruses (AAVs) have shown great potential for human gene therapy. AAV type 2 (AAV2) is the most widely used serotype and was the first AAV used for therapeutic gene delivery. However, vectors based on at least seven other AAV serotypes have also been developed and are being actively used as gene delivery vectors. The interest in these alternative serotypes stems from the fact that they exhibit different cell tropisms. Recently, AAV vector systems have been engineered in an attempt to physically redirect tropism. Many groups have demonstrated that several sites in the AAV2 capsid protein are amenable to insertion of small peptides, and hypothesized that if these peptides have receptor binding specificity, the virus should be able to attach and infect cells through these engineered interactions. Using this approach, AAV-mediated gene delivery can be enhanced or expanded to previously impermissible cell types.
