Cat. No. AD18654Z
Description Fiber modified Ad5/RGD chimeric adenoviral particles which express green fluorescent protein (GFP) reporter gene.
Target Gene GFP
Application Ad5/RGD-GFP chimeric adenoviruses are widely used in a variety of biomedical research applications, including: Gene therapy research: Due to its enhanced infectivity and broad cell tropism, this chimeric virus can serve as an efficient vehicle for delivering therapeutic genes to target cells, making it a valuable tool for developing gene therapy strategies for a variety of diseases. Cancer research: The RGD modification allows for preferential targeting of tumor cells, which often overexpress integrins. This makes the Ad5/RGD-GFP virus a powerful vehicle for oncolytic viral therapy and for studying the interaction of cancer cell biology with the tumor microenvironment. Cell tracking and imaging: GFP expression enables researchers to track the fate of infected cells in real time, facilitating the study of cell migration, proliferation, and differentiation in both in vitro and in vivo models. Vaccination strategies: Viruses can be engineered to express antigens from a variety of pathogens, serving as potential platforms for vaccine development.

Creative Biogene ensures high-quality adenovirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between adenovirus particle lots.
Endotoxin Endotoxins, primarily derived from Gram-negative bacteria, can trigger adverse immune responses. Endotoxin contamination is a significant concern in adenovirus production, especially for applications in animal studies and gene therapy. Creative Biogene utilizes rigorous endotoxin detection methods to monitor the endotoxin level in our produced adenovirus particles to ensure regulatory compliance.
Sterility Creative Biogene ensures that adenovirus products are free of any bacterial, fungal and other microbial contamination.
Ad5 E1 Detection All Creative Biogene adenoviruses are PCR tested to ensure that there are no detectable E1 sequences in the particles, which could be from revertants or external E1 contamination.
RCA Assays Adenovirus products originating at Creative Biogene are guaranteed to have undetectable replication-competent adenovirus (RCA). This quality control measure is important because there is always the possibility of wild-type contamination due to revertants or environmental sources.
PFU Titering All purified adenovirus preparations are tested for infectious titer. Creative Biogene's PFU test takes a few days longer but counts true plaques in HEK cells rather than estimating PFU titers via IHC staining or TCI50 of infected cells.

Customer Reviews

The Ad5/RGD-GFP chimeric adenovirus represents a breakthrough advancement in viral vector technology, specifically designed to improve gene delivery efficiency and cell specificity. This modified adenoviral particle is derived from the common human adenovirus serotype 5 (Ad5) and optimized to express a green fluorescent protein (GFP) reporter gene. A key feature of Ad5/RGD-GFP is its fiber modification. The native Ad5 fiber protein, which is responsible for initial binding to the coxsackievirus and adenovirus receptor (CAR) on host cells, has been engineered to contain an RGD (arginine-glycine-aspartic acid) peptide motif. This RGD modification broadens the tropism of the virus, enabling it to bind to integrins, specifically αvβ3 and αvβ5, which are more widely expressed on various cell types, including cells lacking CAR. This targeting mechanism significantly enhances the ability of the virus to infect a wider range of cells, especially those relevant to cancer and gene therapy research. The integration of a green fluorescent protein (GFP) reporter gene into the viral genome allows for real-time visualization and quantitative analysis of viral infection and gene expression. GFP is a highly stable, easily detectable fluorescent protein that emits bright green fluorescence when exposed to blue light or UV light. Expression of GFP provides researchers with a powerful tool to monitor viral distribution, infection efficiency, and gene expression levels in living cells and tissues using standard fluorescence microscopy or flow cytometry techniques.
