BJ cells are a well-characterized human fibroblast cell line originally derived from normal neonatal foreskin tissue; they are widely used in biomedical research as a representative model of normal human diploid fibroblasts. Because BJ cells retain many physiological characteristics of primary fibroblasts while offering greater consistency and practicality for routine laboratory use, they are frequently selected for studies involving cell growth, senescence, extracellular matrix biology, wound healing responses, cytotoxicity assessment, and cell signaling. The GFP reporter cell line—BJ is an engineered derivative of the BJ cell line, modified to stably express the green fluorescent protein (GFP) reporter gene. Through the stable integration or long-term maintenance of the GFP expression cassette, this reporter cell line provides a reliable fluorescent signal across various passages under appropriate culture conditions.

The GFP reporter cell line—BJ has broad applications in fields such as cell biology, pharmacology, toxicology, and biotechnology. In cell tracking studies, GFP fluorescence enables researchers to monitor the behavior of BJ fibroblasts over time within 2D cultures, 3D matrices, co-culture systems, and tissue-like models. This is particularly valuable for wound healing and migration assays, allowing researchers to observe how fluorescently labeled fibroblasts migrate into scratch wounds, invade extracellular matrix components, or interact with epithelial cells, endothelial cells, immune cells, and tumor cells. In drug screening and cytotoxicity testing, combined with automated imaging or fluorescence intensity measurements, the GFP signal serves as a convenient indicator for assessing viable cell numbers, growth inhibition, or changes induced by drug treatments. As BJ cells are non-tumorigenic human fibroblasts, the BJ-GFP reporter cell line can also be used to evaluate the effects of compounds on normal stromal cells, thereby helping researchers compare selective toxicity between cancer cells and normal fibroblast-like cells.
