NCI-H520 is a well-characterized human lung cancer cell line derived from squamous cell carcinoma of the lung, widely used as an in vitro model for studying non-small cell lung cancer (especially squamous cell carcinoma biology). These cells are typically grown in adherent epithelial-like cultures, providing a useful platform for studying tumor cell proliferation, survival, migration, drug response, signaling pathways, and mechanisms associated with malignant progression. The GFP reporter gene cell line – NCI-H520 is an engineered version of the parental NCI-H520 cell line that stably expresses green fluorescent protein (GFP). Constitutive GFP signaling allows for direct observation and quantification of live tumor cells without additional staining. By combining the disease relevance of the NCI-H520 lung squamous cell carcinoma model with a reliable fluorescent reporter system, this cell line provides a convenient and reproducible tool for fluorescence-based detection. Stable GFP expression enables researchers to monitor cell growth, morphology, distribution, and experimental responses using fluorescence microscopy, live-cell imaging platforms, flow cytometry, fluorescence microplate readers, and high-content imaging systems.

The GFP reporter gene cell line NCI-H520 can be applied to a wide range of cancer research and drug discovery workflows. In oncology research, it can be used to evaluate the effects of anticancer compounds, targeted therapies, combination therapies, and cytotoxic drugs on lung squamous cell carcinoma cells. Because GFP fluorescence is correlated with the presence and relative abundance of labeled cells, this cell line can be used for cell proliferation assays, growth inhibition studies, cell viability-related assays, and kinetic monitoring of drug responses. It is also valuable for high-content screening, enabling the analysis of changes in cell number, morphology, confluence, and spatial distribution after treatment using automated imaging techniques. In cell biology research, GFP-labeled NCI-H520 cells can help researchers observe tumor cell migration, wound healing, invasion, colony formation, and interactions with stromal cells, immune cells, endothelial cells, or extracellular matrix components in co-culture systems. The fluorescence signal can distinguish tumor cells from unlabeled cells in mixed cell populations, which is particularly important for studying tumor microenvironment interactions.
