Safety Considerations The following safety precautions should be observed.
1. Use pipette aids to prevent ingestion and keep aerosols down to a minimum.

3. Wash hands after handling the stable line and before leaving the lab.
4. Decontaminate work surface with disinfectant or 70% ethanol before and after working with stable cells.
5. All waste should be considered hazardous.
6. Dispose of all liquid waste after each experiment and treat with bleach.

Saos-2 is a mature human osteosarcoma cell line derived from primary osteosarcoma tissue, widely used as an in vitro model for bone tumor biology, osteoblast-like differentiation, and bone research. These cells exhibit an epithelial-like morphology, adhere firmly to surfaces treated with standard tissue culture, and are renowned for their osteoblastic characteristics, such as alkaline phosphatase activity, extracellular matrix production, and the ability to form mineralized nodules under appropriate induction conditions. Saos-2 cells are also frequently used in studies of cell cycle regulation, DNA damage responses, apoptosis, and tumor suppressor pathways, particularly because their lack of p53 provides a useful background for investigating p53-related mechanisms. The GFP reporter cell line-Saos-2 is a genetically engineered derivative of the parental Saos-2 cell line that stably expresses green fluorescent protein.
