Creative Biogene ensures high-quality adenovirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between adenovirus particle lots.
Endotoxin Endotoxins, primarily derived from Gram-negative bacteria, can trigger adverse immune responses. Endotoxin contamination is a significant concern in adenovirus production, especially for applications in animal studies and gene therapy. Creative Biogene utilizes rigorous endotoxin detection methods to monitor the endotoxin level in our produced adenovirus particles to ensure regulatory compliance.
Sterility Creative Biogene ensures that adenovirus products are free of any bacterial, fungal and other microbial contamination.
Ad5 E1 Detection All Creative Biogene adenoviruses are PCR tested to ensure that there are no detectable E1 sequences in the particles, which could be from revertants or external E1 contamination.
RCA Assays Adenovirus products originating at Creative Biogene are guaranteed to have undetectable replication-competent adenovirus (RCA). This quality control measure is important because there is always the possibility of wild-type contamination due to revertants or environmental sources.
PFU Titering All purified adenovirus preparations are tested for infectious titer. Creative Biogene's PFU test takes a few days longer but counts true plaques in HEK cells rather than estimating PFU titers via IHC staining or TCI50 of infected cells.

The U6-shRNA-RFP adenovirus is a recombinant adenovirus serotype 5 designed specifically for experimental control applications in molecular biology and gene therapy research. This vector features specific deletions in the E1 and E3 regions, which are crucial for rendering the virus replication-deficient and enhancing its safety. The U6 promoter drives the expression of a random sequence shRNA, designed as a non-targeting control to rule out off-target effects in RNA interference (RNAi) experiments. This shRNA does not silence any specific gene and therefore serves as a baseline reference. In addition, the adenovirus incorporates a reporter system where the red fluorescent protein (RFP) gene is under the control of a strong cytomegalovirus (CMV) promoter. RFP expression allows for convenient visualization and quantification of transduction efficiency in infected cells or tissues. This dual-function design ensures the delivery of a non-functional shRNA control and reliable fluorescent labeling for tracking viral infection and distribution, making it a valuable tool for validating experimental setups.

The U6-shRNA-RFP adenovirus is widely used as a negative control in gene knockdown experiments across various biological models, including in vitro cell culture and in vivo animal studies. In cultured cells, it helps researchers differentiate between specific gene silencing effects and non-specific outcomes caused by viral transduction or shRNA delivery mechanisms. By providing a random sequence shRNA, it controls for potential cellular stress responses, immune activation, or off-target interactions that might confound RNAi assay results. In animal models, the RFP reporter gene allows for real-time monitoring of viral spread, biodistribution, and persistence within tissues, aiding in the assessment of delivery methods and vector tropism.
