Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots.
Mycoplasma Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination.
Purity Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards.
Sterility The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities.
Proviral Identity Confirmation All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert.

The scFv(EGFR)-CD28-OX40-CD3ζ CAR-T lentivirus is a highly advanced gene delivery system designed to modify T cells for targeted immunotherapy. This lentiviral vector is characterized by its efficient and stable transfer of genes to primary human T cells, ensuring the integration of the chimeric antigen receptor (CAR) into the host genome for long-term expression. As a third-generation CAR construct, it possesses a complex intracellular signaling structure, including the major activating domain CD3ζ and two potent co-stimulatory domains CD28 and OX40. The co-inclusion of CD28 and OX40 provides significant advantages over previous generations of CARs; CD28 promotes rapid T cell proliferation and early activation, while OX40 enhances long-term T cell survival, metabolic adaptation, and the formation of memory T cells. This dual co-stimulation significantly improves the persistence and anti-tumor efficacy of CAR-T cells, especially in the complex immunosuppressive microenvironment of solid tumors.

The application of this lentiviral system is primarily focused on the research and development of therapeutic strategies targeting malignancies overexpressing epidermal growth factor receptor (EGFR). EGFR is a key target in oncology because its dysregulation is a hallmark of various aggressive cancers, including glioblastoma, non-small cell lung cancer, and head and neck squamous cell carcinoma. Using these lentiviral particles, researchers can generate highly specific CAR-T cells and evaluate their cytotoxicity against EGFR-positive tumor cells in in vitro experiments and in vivo animal models. In addition to its direct use in therapeutic development, the scFv(EGFR)-CD28-OX40-CD3zeta CAR-T lentivirus is also an important tool for studying mechanisms such as T cell exhaustion, cytokine release syndrome (CRS) models, and the dynamics of T cell infiltration in solid tumors.
