The scFv (CD38)-CD28-CD3zeta CAR-T lentiviral vector is a breakthrough immuno-oncology treatment designed to specifically target and eliminate cancer cells expressing CD38. Chimeric antigen receptor T-cell (CAR-T) therapy utilizes a patient''s own immune cells, engineering them to more effectively recognize and destroy malignant cells. The key component of this CAR-T construct is a single-chain variable fragment (scFv), derived from an antibody that specifically binds to CD38. CD38 is a glycoprotein overexpressed in a variety of hematologic malignancies, including multiple myeloma and various leukemias. The CAR contains not only the scFv but also the costimulatory domains: CD28 and CD3zeta. In addition to the initial signal provided by the scFv binding to its antigen, this domain also provides a second signal required for T cell activation. The inclusion of the CD3zeta signaling domain is crucial for delivering the activation signal to T cells, thereby initiating a potent immune response against cancer cells. This dual signaling mechanism is crucial for achieving optimal T cell activation, enhancing cytotoxicity, and promoting sustained anti-tumor responses.
