Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots.
Mycoplasma Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination.
Purity Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards.
Sterility The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities.
Proviral Identity Confirmation All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert.

The scFv (CD38)-CD28-CD3zeta CAR-T lentiviral vector is a breakthrough immuno-oncology treatment designed to specifically target and eliminate cancer cells expressing CD38. Chimeric antigen receptor T-cell (CAR-T) therapy utilizes a patient''s own immune cells, engineering them to more effectively recognize and destroy malignant cells. The key component of this CAR-T construct is a single-chain variable fragment (scFv), derived from an antibody that specifically binds to CD38. CD38 is a glycoprotein overexpressed in a variety of hematologic malignancies, including multiple myeloma and various leukemias. By utilizing the scFv, this CAR-T therapy is designed to achieve high specificity and affinity for CD38-expressing tumor cells, minimize off-target effects, and protect normal cells.

The CAR contains not only the scFv but also the costimulatory domains: CD28 and CD3zeta. The CD28 costimulatory domain plays a crucial role in enhancing T cell activation, proliferation, and longevity. In addition to the initial signal provided by the scFv binding to its antigen, this domain also provides a second signal required for T cell activation. The inclusion of the CD3zeta signaling domain is crucial for delivering the activation signal to T cells, thereby initiating a potent immune response against cancer cells. This dual signaling mechanism is crucial for achieving optimal T cell activation, enhancing cytotoxicity, and promoting sustained anti-tumor responses.
